Design Notes

This sRNA has an antisense region of 20 nucleotides. We also constructed two other sRNA that has an antisense region specific for USP45. The difference is the length of the antisense region. One has an antisense region of ten nucleotides and the other has one with 15 nucleotides. Usually there is a trade of in specificity and strength with varying length of antisense region. And that’s the reason we have three different sRNA. These three parts should work in theory but have not been sufficiently tested and the relative strength of the three parts is unknown. But we know that it doesn’t bind to any vital gene in e.coli and silence that because it was possible to express this silencing sRNA.
